Multiple Applications for Protein Functional Analysis–More Versatile than GFP

With HaloTag® Technology you can perform multicolor live- and fixed-cell imaging experiments using a single genetic construct. The technology is based on the formation of a covalent bond between a protein fusion tag and synthetic ligands, and is designed to enable understanding of protein function in a cellular and biochemical environment.

Fluorophores attached to HaloTag® ligands with different abilities to cross the cell membrane are used to regionally label a protein of interest.

Multiplex Live-Cell Imaging

The HaloTag® protein is not intrinsically fluorescent (unlike GFP). Therefore, the choice of fluorescent labels, including secondary and tertiary fluorophores, can be made after creating the HaloTag® fusion protein. This gives you more flexibility in choice of fluorophores than GFP.

The HaloTag® protein-ligand interaction is stable and highly specific. The fluorescently labeled HaloTag® protein can be boiled with SDS-PAGE sample buffer and resolved on a gel without loss of fluorescent signal.
